COMPOSITION:
Each 2mL ampoule contains:
Gentamicin      80 mg.
Excipients:         q.s.
PHARMACOLOGICAL:
Gentamicin is an aminoglycoside antibiotic which is active against many strains of gram-negative bacteria including pseudomonas and against many Staphylococcus aureus strains Enterocoel and streptococci are insensitive to gentamicin alone, but it exhibits synergy with beta lactams against these organisms.
PHARMACOKINETICS:
This product is absorbed rapidly after intramuscular injection and completely, in the 0.5 - 1 hours to reach Cmax. T½ is 2 - 3 hours. Renal dysfunction can be significantly increased.The low rate of protein binding in the body can be distributed in a variety of organizations and body fluids. in the accumulation of renal cortical cells can enter the fetus through the placental barrier in vivo, it is not easy through the blood - brain barrier into the brain tissue and cerebrospinal fluid.
Metabolism in the body not to prototype. With the urine by glomerular filtration discharged within 24 hours after delivery from the dosage of 50% - 93%.
INDICATIONS:
For the treatment of Urinary tract infections, septicemia due to Gram-negative Bacilli, Bums, Pediatrics Infections, Chemoprophylaxis in abdominal surgery, Biliary tract infections, other infections due to susceptible organisms.
CONTRA-INDICATIONS:
Hypersensitivity to gentamicin is contraindication to its use. A history of hypersensitivity or serious toxic reactions to other aminoglycosides may contraindicate use of gentamicin because of the known cross sensitivity of patients to drugs in this class.
PRECAUTIONS/WARNINGS:
1. Cross-allergy of aminoglycoside antibiotics in patients with allergies may be allergic to gentamicin.
2. The following circumstances shall be used with caution: water loss, the first eight pairs of cranial nerve damage, myasthenia gravis or Parkinson's disease, patients with renal dysfunction.
3. Patients should be given adequate water, so as to reduce the renal tubular damage.
4. Long-term application may lead to excessive growth of resistant bacteria.
5. Should not be used for subcutaneous injection.
6. Gentamicin should be used for caution with renal impairment, myasthenia gravis, Pregnancy, Elderly.
INTERACTIONS:
Increased nephrotoxicity with cephalosporins, amphotericin, cyclosporine, mannitol, plasma substitutes.
Increased ototoxicity with frusemide, other aminoglycosides. Prolongs effects of neuromuscular-blocking drugs & anesthetics.
ADVERSE EFFECTS:
The main side effects are ototoxicity and monitoring of plasma concentration is important to avoid toxic concentrations. Other adverse effects include neuromuscular blockade, hypersensitivity blood dyspraxias and central and peripheral neurotoxicity.
ADMINISTRATION:
Gentamicin injectable is given by I.M. or I.V. route. Or as directed by the physician. 
ADULTS:
In the treatment of urinary tract infection 0.8 to 1.2 mg/kg body weight is administered I.M. every 8 hourly in patients with normal kidney functions.
Patients with damaged kidney should be given more than 1 mg gentamicin and the injections may be given at longer intervals.
For systemic infection, the recommended dosage of Gentamicin in 3 - 5 mg/kg administered in three equal doses every 8 hours.
If the infection is life threatening, dosage up to 5 mg/kg/day maybe administered and divided doses. These doses should be reduced to 3 mg/kg/day as soon as clinically indicated.
CHILDREN:
2 to 2.5 mg/kg administered every 8 hours.

INFANTS AND NEONATES: 
2.5 mg/kg administered every 8 hours. Premature or full-term neonates one week of age or less 2.5 mg/kg administered every 12 hours.

Ampoules of 2ml-box of 10 ampoules.
STORAGE:
Store below 25ºC. Protect from light.
KEEP OUT OF REACH OF CHILDREN.
